Can I use the Honeywell 69-2384ES-01 RTH111 with a heat pump system?

Yes. Set jumper JP2 on the back of the thermostat to match your heat pump wiring: leave it in the factory position if you've connected O wire to the O/B terminal, or move it to position B if you've connected B wire to that terminal. Installation Guide, p. 10

Where is the locking screw on this thermostat?

At the bottom of the thermostat. It's captive and can't be removed from the wallplate. Installation Guide, p. 6

What does the C terminal mean on the Honeywell 69-2384ES-01?

C and C1 terminals are not used on this thermostat. If wires are attached to them on your old unit, wrap the bare metal ends with electrical tape and don't connect them to the new thermostat. Installation Guide, p. 5
